- She started playing tennis at the age of 4 with the guidance of her father. At the age of 10, she started practicing in Prague. At the age of 15, she moved to Prague all by herself to train.
- She has a tattoo situated above her right elbow that reads, “No rain, no flowers”.
- Vondroušová made her WTA-Tour main-draw debut in doubles at the 2015 Sparta Prague Open with her doubles partner Kateřina Vaňková. They lost the first round to Kateryna Bondarenko and Eva Hrdinová.
- Vondroušová made her WTA debut in singles at the 2016 J&T Banka Prague Open during which she defeated Océane Dodin before her second-round loss to eventual tournament runner-up Samantha Stosur. She then played at the Roland Garros junior event but lost in the 3rd round to Olesya Pervushina. After 2 consecutive losses, she went on a hiatus for the rest of the year to recover from an elbow injury.
- Following her comeback to the 2017 tour, she was quick to compile 20 consecutive wins which resulted in one $15,000 title, one $25,000 title, and 2 more $25,000 finals.
- In April 2017, she reached her debut WTA singles final at the Ladies Open Biel/Bienne, scoring wins over Annika Beck, , and . Following her multiple winning streaks, she got past Anett Kontaveit in the final to clinch her maiden WTA title, which brought her close to a top-100 debut, making her the youngest winner of a WTA title since Konjuh’s title at the 2015 Nottingham Open.
- In 2017, she made her Fed Cup debut in the semifinals in a match against the United States but lost to Coco Vandeweghe in straight sets. In her 2nd match, she won her first Fed Cup in a match against Lauren Davis in straight sets.
- She reached the women’s doubles semifinal of the Australian alongside her doubles partner Barbora Strýcová in January 2019 but lost the match to Sam Stosur and Zhang Shuai. In singles, she defeated Evgeniya Rodina in the first round but lost to Petra Martić in the 2nd round. She reached the final round in Budapest, beating Georgina Garcia Perez, Anna Blinkova, Irina-Camelia Begu, and Anastasia Potapova. After defeating a row of competitors on her way to finals, she then lost to Alison Van Uytvanck.
- In the 2019 French Open, she became the first teenager to make it to the finals and the first teenager to reach a Grand Slam finals. Though she had multiple winning streaks until the semifinals where she defeated , she lost the final match in straight sets to . After she encountered problems with her left wrist, she underwent surgery and was sidelined for the rest of the season.
